dc.description.abstractPlectranthus edulis (Vatke) Agnew (syn. Coleus edulis Vatke), is a diploid, dicotyledonous plant, belonging to the Labiateae (Lamiaceae) family. It is a major source of food in many parts of Ethiopia and also used as a traditional medicine to treat different diseases. This plant is vegetativelly propagated mainly by tubers. Vegetativelly propagated plants are mostly affected by different diseases. These diseases reduce the yield and results in poor quality crops. The establishment of in vitro plant regeneration system in P. eduils is of potential importance to quality improvement and genetic transformation. The study was conducted to develop in vitro regeneration protocol for P. eduils. For this study, calli were induced from leaf explants excised from three-week-old shoots grown on MS medium containing different concentrations of NAA and BAP. The highest (100 %) callus induction was obtained on MS medium supplemented with 1.5 mg/l NAA+1.0 mg/l BAP and 2 mg/l NAA+0.5 mg/l BAP. Among the different growth regulators combinations used for multiplication, maximum (58.68±6.57) mean numbers of shoots were obtained on 0.5 mg/l BAP in combination with 0.4 mg/l GA3. In vitro developed shoots were rooted best with mean root number of 10.15±0.95 on MS medium supplemented with 2.0 mg/l IBA. All in vitro rooted and 78 % of microshoots were survived in the glasshouse. This protocol can be used to improve this plant through in vitro screening and genetic transformation.en_US
