Tesfaye SissayGeJagay AyeJetWossene Negash2026-08-042026-08-042011-06https://etd.aau.edu.et/handle/123456789/8751This study was conducted from September 20 10- June 20 11 at Debre Zeit National Veterinary Institute (NVI) with an attempt to isolate and characterize respiratory viruses from pneumonic lungs of camels slaughtered at Addis Ababa Akaki abattoir. Accordingly, pneumonic camel lung tissues were collected and transported in ice box to NVI virology laboratory and temporarily stored at -85°C and then processed and inoculated on confluent vero cells mono layers, incubated at 37°C and followed for the development of CPE for 4-15 days. All CPEs observed developed between 4-14 days. Thirty five (35) pneumonic camel lungs were cultured among which 27 of them were CPE positive characterized majorly by initial rounding of vero cells, elongation, syncytia formation, swelling and rounding, floating and sloughing of cells. Some of the supernatant suspensions of those samples exhibiting CPEs were taken and extracted both for DNA and RNA viral genomes. cDNAs were synthesized from RNA extracted samples. The DNAs and cDNAs were used to test both DNA and RNA viruses, respectively using universal (DOP- PCR) and conventional PCR techniques. Respiratory syncytial virus, Adenovirus, Pest des polities ruminants virus, Parainfluenza I virus, Parainfluenza 2 virus and Parainfluenza 3 virus were tested. All six samples tested were negative for PPRV and Parainfluenza virusesl-3. But nine of ten samples tested were positive for Respiratory syncytial virus and two of six samples tested were positive for Adenovirus. Finally, further investigative, epidemiological and molecular characterization studies were forwarded .
